Application of phosphate buffer
Phosphate buffered saline (PBS), the most widely used buffer in biochemical research, the main components are Na2HPO4, KH2PO4, NaCl and KCl, generally used as a solvent, as a dissolution protection reagent. Due to the secondary dissociation of Na2HPO4 and KH2PO4, the buffered pH range is wide, while NaCl and KCl mainly increase the salt ion concentration. If required, PBS can also be supplemented with 1 mmol/L CaCl2 and 0.5 mmol/L MgCl2 to provide divalent cations.
PBS is the abbreviation of phosphate buffered salt solution, not only pseudorabies virus to be diluted with it, but also the general active biological agents to use it dilution.
The reason is that it has salt balance, adjustable suitable pH buffering, distilled water does not have salt balance, can destroy the structure and biological characteristics of biological proteins; Saline does not have the effect of adjusting pH, and there is no guarantee that intact, active substances will participate in biological reactions under optimal conditions, so PBS is preferred. PBS is not a panacea, and some bioactive substances require higher conditions than PBS, which requires adding more components to the equilibrium buffer to maintain the best conditions to ensure that the bioactive substances maintain their most complete characteristics
